Choosing a diluent
The choice is usually between bacteriostatic and preservative-free sterile water. Bacteriostatic water suits multi-draw work because the benzyl alcohol limits microbial growth; preservative-free water suits single-use preparations and any assay where benzyl alcohol is itself a confound.
Volume and concentration
Concentration follows from the mass in the vial and the volume added, so the volume has to be measured rather than estimated. Recording it at the point of reconstitution is what makes results comparable between vials of the same lot.
Storage after reconstitution
Reconstituted material degrades considerably faster than lyophilized powder. Refrigerate, protect from light, and work to whatever window the protocol specifies rather than to the dry-powder shelf life.

- Sterile water containing 0.9% benzyl alcohol, which inhibits microbial growth and so permits repeated withdrawals from one vial.
- Sterile water for injection
- Preservative-free sterile water, appropriate where a preservative would interfere with the assay — but suited to single use only.

Handling and storage
Add diluent slowly down the inside wall of the vial rather than directly onto the powder, and allow it to dissolve without agitation — shaking can denature a peptide. Record the volume added, since concentration is otherwise not reproducible across a batch.
